چکیده

LEVEL: INTERMEDIATE T he topology of plasmid DNA can be monitored with capillary gel electrophoresis (CGE) (1, 2). An improved CGE technology was developed at PlasmidFactory to routinely determine the content of different plasmid forms that appear during cultivation and purification in DNA production runs. CGE is a useful tool for supporting process development in plasmid DNA and minicircle production, in drug delivery research, and in plasmid storage and stability evaluation studies. Long-term storage of the lacZ (beta-galactosidase)–expressing pCMVß plasmid over a period of 13 months revealed that stable storage conditions at –80 °C (rather than storage at 4 °C) prevent an increase in open circular (oc) plasmid topology deriving from the degraded, supercoiled, covalently closed circular (ccc) form. Physical data were supported by in vivo gene transfer data, using jet-injection into different tumor models grown subcutaneously in mice. Additionally, plasmid pCMV-S2S produced for clinical applications has been stored at –20° C and monitored for more than five years with no detectable decrease of quality (no topology changes).

A seven-year storage report of good manufacturing practice-grade naked plasmid DNA: stability, topology, and in vitro/in vivo functional analysis.

The great interest for naked plasmid DNA in gene therapy studies is reflected by the fact that it is currently used in 18% of all gene therapy trials. Therefore, validation of topology and functionality of DNA resulting from its long-term stability is an essential requirement for safe and effective gene transfer. To this aim, we analyzed the stability of good manufacturing practice-grade pCMVβ ...

Plasmid DNA replication and topology as visualized by two-dimensional agarose gel electrophoresis.

During the last 20 years, two-dimensional agarose gel electrophoresis combined with other techniques such as Polymerase Chain Reaction, helicase assay and electron microscopy, helped to characterize plasmid DNA replication and topology. Here we describe some of the most important findings that were made using this method including the characterization of uni-directional replication, replication...

Restriction endonuclease analysis of nosocomial isolates of Clostridium difficile.

A total of 110 clinical isolates of Clostridium difficile were analyzed by agarose gel electrophoresis by using both bacterial restriction endonuclease analysis (REA) and plasmid profiles. A total of 72 isolates were divided into 12 groups according to their REA patterns. Some 38 isolates exhibited unique patterns. Pattern A occurred in 20% of isolates. Isolates with patterns B, E, and G were c...

Optimization of the Analysis of Almond DNA Simple Sequence Repeats (SSRs) Through Submarine Electrophoresis Using Different Agaroses and Staining Protocols

Simple sequence repeat (SSR markers or microsatellites), based on the specific PCR amplification of DNA sequences, are becoming the markers of choice for molecular characterization of a wide range of plants because of their high polymorphism, abundance, and codominant inheritance. Different methods have been used for the analysis of the SSR amplified fragments being submarine agarose electropho...

Construction of Mtb72F Plasmid as a DNA Vaccine Candidate for Mycobacterium tuberculosis

Background:  With one-third of the world’s population infected, tuberculosis (TB) is one of the most common infectious diseases and a major public health problem, especially in developing countries. The efficacy of the BCG vaccine for controlling the disease in adults is poor. The development of an effective TB vaccine is a global objective. An effective tuberculosis vaccine should s...
